چکیده

The global effort to understand the molecular drivers of cancer onset and progression is now coming to fruition with the identification of specific genomic and epigenomic events that influence signaling through key oncogenic pathways. Genetic studies using inducible expression of the critical growth controlling oncogenes MYC, RAS, PI3K and AKT have shown unequivocally that, in conjunction with secondary genetic mutations, they can drive transformation and induce oncogene addiction. Since the majority of human tumors exhibit dysregulated signaling via one or more of these pathways, this provides an unprecedented opportunity to improve patient outcome by therapeutically targeting this addiction. These key drivers of malignant transformation do so by controlling many of the processes regarded as hallmarks of cancer, including proliferative cell growth and resistance to apoptosis, reprogramming of energy metabolism, angiogenesis and metastasis. Paradoxically, however, the MYC, RAS and PI3K/AKT oncogenic pathways can also induce cellular senescence in non-transformed cells. Pandolfi and colleagues proposed that RAS oncogene-induced senescence (OIS) differs from loss of the tumor suppressor PTENinduced cellular senescence (PICS) by the absence of a DNA damage response, and our publication demonstrated oncogenic AKT-induced senescence similar to PICS. In all three cases, senescence relies on modulating p53 levels and activity and/or INK4A mediated inhibition of cyclin-dependent kinases (CDKs) and, thus, inactivation of the retinoblastoma Too much or too little Harnessing senescence to control oncogene-driven cancer

Bridging the Gap Between Research and Policy and Practice; Comment on “CIHR Health System Impact Fellows: Reflections on ‘Driving Change’ Within the Health System”

Far too often, there is a gap between research and policy and practice. Too much research is undertaken with little relevance to real life problems or its reported in ways that are obscure and impenetrable. At the same time, many policies are developed and implemented but are untouched by, or even contrary to evidence. An accompanying paper describes an innovative progr...

Do people trust too much or too little?

Across two studies, we asked whether people trust too much or too little, relative to what an economic analysis would suggest. In the trust game paradigm, participants decided whether to hand money over to an anonymous individual who could either return more money back or keep all the money. Participants trusted too little, in that they grossly underestimated the proportion of their peers who w...

This article attempts to focus on national media audience on one hand and satellite television net...

From thinking too little to thinking too much: a continuum of decision making.

Due to the sheer number and variety of decisions that people make in their everyday lives-from choosing yogurts to choosing religions to choosing spouses-research in judgment and decision making has taken many forms. We suggest, however, that much of this research has been conducted under two broad rubrics: The study of thinking too little (as with the literature on heuristics and biases), and ...
